Missing on Cylinder #1

Well the new 289 is missing on #1 checked the plug and it was fine, changed the dizzy cap and same problem. My dad thinks the valves are too tight on the #1 so we are gonna check that. Also we only have vacuum in neutral for the power brakes so were not sure about that. Thanks guys Oh, we changed plug wires, and made sure we had the right firing order.
 
Vacuum for the poer brake should be hooked up to a full manifold vacuum source, like on the back of your carb or on the manifold itself. If it is, then it sounds like you have a major vacuum leak and some of the plugs will miss
 
after further investigation, several of the cylinders have no effect when the plug wire is removed. I bought this manifold used which was probably the wrong thing to do. It was painted when I got it which made me slightly suspicious to begin with. One more thing, The carb an edlebrock 650cfm 4 barrel has two screws for mixture on it, when running the screw on the left has absolutely no effect. Ideas?
