Depends on what he wants to do with the car. If he is going to rev it up and see that 485 hp constantly then the chances of going BOOM will be greater. If he is just using it for street and wont see full power all the time then it will last longer. The general concensus is that past 450, it's just a matter of time before the rods let go. Also, how high he revs it past 6000 plays a factor. 485 at 6000 and 485 at 6600 are two different things. For every 10% you raise the RPM, the stress goes up 50% at the same power level.

As far as trannies go, if he sees a lot of track time i.e. launching hard on sticky tires a lot, he will probably want to upgrade is input shaft and clutch. Other than that it seems the tranny is good for 500hp.

If he does want to go to 500, he should probably do the rods and pistons at a minimum. Some people will say you might as well do the crank to but others will say the stock crank is fine.

A custom tune will always be better than off the shelf "canned". Espcially if you are not running 100% the same spec as the canned tune.

I agreeing running high HP and beating on the car is more likely to break stuff sooner. If they truly want a sustanable & reliable ride with 500rwhp then building the motor is a must. But will also be costly.
