Will my existing hot air choke tube coming off the exhaust manifold hook up to the Autolite 4100 without any modifications? Only if I use a vintage factory 4-barrel intake?

The 302 hot air choke did not involve the intake manifold, the heat source is the RH exhaust manifold. The tubes used were identical either way. If you use an aftermarket manifold, the slight relocation of the choke will be insignificant, and the tubes will hook up as though stock.
Good choice on the carb, BTW. The Autolite 4100 is probably the best 4V carb ever made. The only limitation is they were never made beyond a nominal 600 cfm (actually a little smaller) except for an ultra-rare one year only 650.
The Summit™ carburetor was Holley's attempt, about 20 years ago, to copy some of the 4100 features, such as annular discharge (only on the primaries) and top cover on a one-piece bowl and body. It had a few things going against it- Not a traditional Holley design,not well marketed, and they didn't copy enough of the 4100 features. For instance, it had no internal feed for the rear bowl, and required a bulky external dual fuel line, like a Holley double-pumper.
